Product Information

Product Usage Information

Application Dilution
Western Blotting 1:1000
Immunofluorescence (Immunocytochemistry) 1:400 - 1:1600

Storage

Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/mL BSA, 50% glycerol, and less than 0.02% sodium azide. Store at –20°C. Do not aliquot the antibody.

Specificity / Sensitivity

Desmoglein 3 (5G11) Mouse mAb recognizes endogenous levels of total desmoglein 3 protein. This antibody does not cross-react with desmoglein 1 or desmoglein 2 proteins.

Species Reactivity:

Human

Source / Purification

Monoclonal antibody is produced by immunizing animals with recombinant protein specific to the sequence of human desmoglein 3 protein.

Background

Desmogleins are calcium-binding transmembrane glycoprotein components of desmosomes, which are intercellular junctions that mediate strong cell-cell adhesion and are linked to the intermediate filament cytoskeleton. Dysregulation of desmosomal protein expression affects both cell adhesion and communication, and has been shown to result in various human pathologies (1). Desmoglein 3 (DSG3) expression is altered in some human cancers, and its expression may be linked to tumor progression (2). Auto-antibodies against desmoglein 1 and desmoglein 3 cause Pemphigus, a life-threatening autoimmune disorder that results in blistering of the skin and mucous membranes (3).

Species Reactivity

Species reactivity is determined by testing in at least one approved application (e.g., western blot).

Western Blot Buffer

IMPORTANT: For western blots, incubate membrane with diluted primary antibody in 5% w/v BSA, 1X TBS, 0.1% Tween® 20 at 4°C with gentle shaking, overnight.

Western Blotting Image 1: Desmoglein 3 (5G11) Mouse mAb Expand Image
Western blot analysis of extracts from 293T cells, mock transfected (-) or transfected with a construct expressing Myc/DDK-tagged full-length human desmoglein 3 (hDesmoglein 3-Myc/DDK; +), using Desmoglein 3 (5G11) Mouse mAb (upper), Myc-Tag (71D10) Rabbit mAb #2278 (middle), or GAPDH (D16H11) XP® Rabbit mAb #5174 (lower).
Western Blotting Image 2: Desmoglein 3 (5G11) Mouse mAb Expand Image
Western blot analysis of extracts from various cell lines using Desmoglein 3 (5G11) Mouse mAb (upper) or GAPDH (D16H11) XP® Rabbit mAb #5174 (lower). Low expression of desmoglein 3 protein in A549, Hep G2, HEC-1-A, and SK-MEL-28 cells is consistent with the predicted expression pattern.
